검체 0.5 g에 탄산가스를 함유하지 않은 물 50 mL를 가하고 페놀프탈레인 시액 0.5 mL를 가하여 0.1 N 수산화나트륨용액으로 30초간 홍색이 지속될 때까지 적정한다.
